The recommended chlorambucil dosage is usually between 4 mg and 10 mg each day, depending on your weight, the reason you are taking the drug, and other factors. This prescription drug comes as a tablet and is taken once daily. Try to take chlorambucil at the same time each day to help keep an even level of the medication in your bloodstream.

An Introduction to Your Dosage of Chlorambucil

The dose of chlorambucil (Leukeran®) your healthcare provider recommends will vary, based on a number of factors, including:
  • Your weight
  • The reason you are taking it
  • How you respond to the medication
  • Other medical conditions you may have.
As is always the case, do not adjust your dosage unless your healthcare provider specifically tells you to do so.

Chlorambucil Dosing Guidelines

The recommended initial dosage of chlorambucil for the treatment of lymphoma and chronic lymphocytic leukemia (CLL) is usually 0.1 mg to 0.2 mg per kg body weight (about 0.05 mg to 0.09 mg per lb) each day for three to six weeks. This usually comes out to 4 mg to 10 mg a day.
In general, people with Hodgkin's lymphoma usually require higher doses of about 0.2 mg per kg body weight each day, whereas people with CLL or non-Hodgkin's lymphoma require 0.1 mg per kg each day.
If treatment is to be ongoing, lower doses will be used. The usual recommended maintenance dose of chlorambucil should not exceed 0.1 mg per kg each day, and may be as low as 0.03 mg per kg per day. Typical doses for maintenance treatment are normally 2 mg to 4 mg a day.
Your healthcare provider will monitor your blood counts closely during treatment, and may recommend a lower dosage if your blood counts drop too low. Your healthcare provider may also recommend a lower dosage if you have kidney or liver disease.
Some people may take "pulse" doses of chlorambucil, which means they will take the medicine every other week or once a month, instead of every day. This usual recommended initial chlorambucil pulse dosage is 0.4 mg per kg body weight (about 0.18 mg per lb) every other week or once a month. Doses may be increased by 0.1 mg per kg body weight until the cancer is adequately controlled, or serious side effects occur.
